Abstract: Objective To study the dynamic variations of the effect of saikosaponin a (SSa) on epileptic mice induced by pentylenetetrazol(PTZ),and to understand the effect characteristics of SSa. Method With the epileptic mice induced by PTZ as animal model. In the different time point,the tentative seizure incubation period and the case of tetanic convulsion were recorded to study the characteristics of the antiepileptic effect of SSa on mouse induced by PTZ. Results In the group of 30min,60 min and 90 min,the incubation period of clonic cramp was significantly prolonged by SSa (P0.05),and in the group of 90 min,the effect was remarkable (P0.01). Also,intraperitoneal injection of SSa could prolong the incubation period of tetanic convulsion in the group of 60 min,90 min and 120 min (P0.05).The effect was especially evident in the group of 90 min (P0.01). In the group of 90 min,the transition time was remarkably prolonged by SSa (P0.05). In the group of SSa 30 min and SSa 60 min,SSa could reduce the tetanic convulsion of epileptic mice introduced by PTZ (P0.05),and the effect is more significant in the group of 60min than that in the group of 30min. In the group of 30 min and 60 min,SSa could protect the epileptic mice introduced by PTZ from tetanic convulsion more significantly than those in the group of 90 min and 120 min.Conclusions SSa can inhibit the epileptic mice induced by PTZ,SSa can protect the mouse from tetanic convulsion,and in 2 hours,the effect was more remarkable at 60th min,90th min than others.

柴胡皂苷(saikosaponin)为中药柴胡的主要成分之一,其单体成分之一的柴胡皂苷a(SSa)有降血脂、抗渗出、抗肉芽肿和保护肝细胞等作用[1],对于慢性肾病的治疗也有较好疗效[2,3],但目前尚没有SSa抗癫痫作用的报道。不过已有不少证据表明柴胡及柴胡总皂苷具有镇静和抗实验性癫痫的作用[4,5]。在柴胡皂苷单体各成分中,为明确何种皂苷单体具有抗癫痫作用,笔者研究了SSa对PTZ致痫小鼠痫性发作的影响,并探讨SSa对痫性发作影响的不同时间的动态变化,以期了解SSa抗癫痫药效作用的时间特点。
1 材 料
1.1 动物
健康昆明种小鼠180只,雌雄各半,体质量(20±2)g,由南方医科大学动物实验中心提供,合格证号为2004A063。
